Depends how tall you are! For those taller than around 6ft I’m pretty sure the bed on train will be too short. We took bus for this reason but it was during the afternoon so we didn’t lose sleep. It was fun and a great experience…crazy roads at times but arrived safely. We used Hk Busline and booked the lower cabins (preferred over top ones in case of motion sickness 😉)

What about toilets? What happens on a bus? Do they stop from time to time?

The problem with the overnights is you arrive at 6am and then what do you do? Where do you go at 6am? I would just do a day bus.
